United boss Gary McAllister kept the same line-up that had played so well at Derby County in the Carling Cup four days ago.
United had dominated the Championship side without reward and McAllister was looking for a similar performance against Huddersfield.
The game attracted the biggest crowd of the season to Elland Road, although there were some spaces in the East Stand Lower, and in the away section with Huddersfield failing to sell their full allocation.
The big turnout was rewarded inside three minutes as United took a quick-fire lead, Luciano Becchio delivered a cross from the left and Rob Snodgrass got the decisive touch at the far post.
Huddersfield re-grouped quickly after the goal and, after winning a free-kick on the edge of the area, they also had an early effort which went over the top.
In reply, Fabian Delph lauched a shot wide from distance and Becchio was denied by MIchael Collins on the edge of the box after good play by Ben Parker.
United had the ball in the net again on 20 minutes after a great finish from Andy Robinson, but the effort was ruled out for offside.
Huddersfield returned the free-kick down-field and Gary Roberts tried his luck from distance. Ian Craney also got himself into a good position, but Casper Ankergren read the situation well and averted the danger.
The visitors worked hard to stifle Leeds, and Nathan Clarke became the first booking of the game for a lunge on Snodgrass.
The Scot was a threat for United again, ands he went close on 40 minutes when he curled shot over the bar.
But just as United got off to a quick-fire start in the first half, Huddersfield got themselves back in the contest just 35 seconds into the second half.
The Leeds defence failed to deal with a cross from the right and Joe Skarz hammered home the equaliser.
United hit back straight away and Parker had a shot cleared off the line after working himself an opening.
But Huddersfield were showing plenty of enterprise and Keigan Parker shot over the top from a tight angle after Roberts won the ball in the middle of the park.
Shortly after the hour-mark, McAllister introduced Jermaine Beckford to the action in place of Neil Kilkenny.
And the striker almost made an immediate impact when Matt Glennon made a superb save to deny him with well-placed shot from distance.
On 71 minutes, it was Becchio who went close when he rose well to head a Snodgrass cross against the woodwork.
Jonny Howson and David Prutton joined the fray on 76 minutes as United looked to re-shuffle in the hope of forcing a winner.
On 80 minutes United had what appeared to be a good claim for a penalty waved away when Beckford claimed a push after he was felled in the area.
United were certainly starting to pile on the pressure and after Richardson had a shot blocked, Robinson played in a cross which eluded everyone in the Huddersfield box.
In the 89th minute Robinson curled a shot wide of the mark, but the real drama came in stoppage time when Huddersfield stole a winner.
Former United striker Danny Cadamarteri broke down the right, and Collins was on hand to convert from close range to settle matters.
