United boss Simon Grayson made two enforced changes to his starting line-up with Aidy White and Leigh Bromby coming in for injured duo Shane Lowry and Richard Naylor. New recruit Paul Dickov was also named among the subs for the first time.
The previous meeting between the two sides was a dour 0-0 draw in December, but United immediately went on the front foot in this encounter, Robert Snodgrass forcing Wojciech Sczcesny into a good early save.
Snodgrass also found himself in the book early on following a clash with Ryan Dickson.
Leeds were looking to make the early running, but the opposition had also adopted a more expansive approach than in the previous meeting.
And it was Casper Ankergren who came racing off his line on the half-hour to deny Sam Wood after a good move by the visitors.
United responded immediately, Jermaine Beckford scraping the wrong side of the upright with a header following a great cross from Snodgrass. Moments later, the Scot screwed a shot wide after a neat pass from Jonny Howson.
Eight minutes from half-time, an advancing Sczcesny made a decent stop, reaching low to his left as Beckford looked to angle his shot inside the far post.
Beckford went close again before half-time when he had a shot deflected wide and the striker was also involved when Gary McSheffrey lifted a shot over the top.
Leeds had slowly started to control of the game as half-time loomed, creating chances and playing some good football.
Snodgrass was next to try his luck when he had a shot blocked, but the goal wouldn't come before the half-time whistle sounded.
United started the second half in similar vein - Howson was involved as Beckford tested Sczcesny from distance - but Grayson was forced into an early re-shuffle with Lubomir Michalik replacing an injured Andrew Hughes.
And Leeds were rocked before the hour-mark when Brentford took the lead. Charlie MacDonald returned a cross across the box and with the aid of a deflection the ball sat up for Ben Strevens who headed home from close range.
Grayson's men didn't deserve to be trailing and came straight back at Brentford, Snodgrass sending a shot wide of the mark.
Sczcesny was also called upon to save from his own player, Leon Legge, who headed towards goal when under pressure from Paddy Kisnorbo. Next up, Beckford headed wide following a great run by McSheffrey.
The pressure continued and Leeds were deservedly level on 72 minutes when McSheffrey was again the provider and Beckford steered the ball home from close range.
United immediately went on the hunt for a winner and Snodgrass was crowded out after good work by Beckford, and with 10 minutes remaining Grayson made a double change, introducing Bradley Johnson and Dickov.
As the game headed towards it final minutes, Snodgrass was made a great run and Howson had a shot blocked. Dickov also had a shot blocked and in the 90th minute Michalik hit the inside of the post with a header and the ball bounced agonisingly to safety when Beckford was ready to pounce.
As the game entered stoppage time, Leeds looked the only side likely to take all three points, but the last chance came with a goalmouth scramble that ended when Bromby fired over.
